Influence of Age and of Dietary Restriction on the Mouse Aorta1

Abstract

This study was designed to examine morphological changes in cross sections of the thoracic aorta of male Swiss albino mice during the course of aging. Histometric measurements made on the aortas of mice at 1, 3, 10 and 17.5 months of age showed that the tunica media, which consists of elastin lamellae and smooth muscle layers, thickens significantly with advancing age. To evaluate the effects of dietary restriction on the aorta, the aortas of 20 month old mice on a 4% protein diet (restricted) were compared with those on a 26% protein diet (control). The aortas of the 26% protein fed mice were significantly thicker and the number of elastin lamellae was significantly greater than those of the 4% protein fed mice. The control mice showed a greater degree of age related changes in the aorta than did mice on the protein restricted diet.

Résumé

Dans cette étude, nous examinons les modifications morphologiques sur les sections transverales de l'aorte thoracique de souris mâles “Swiss albino” pendant le processus de vieillissement. Des mesures histométriques de l'aorte de souris âgées de 1, 3, 10 et 17.5 mois démontrèrent que la media, constituée de lamelles élastiques et de muscles lisses, s'épaissit avec l'âge.

Afin d'évaluer les effets sur l'aorte d'une diète limitée en protéine, les aortes de souris âgées de 20 mois et nourries d'une diète de 4% en protéine furent comparées à celles de souris alimentées d'une diète de 26% en protéine (contrôle). L'épaisseur et le nombre de lamelles élastiques des aortes de souris contrôles s'avérèrent significativement plus grands que chez les souris alimentées d'une diète à 4% en protéine. Les aortes de souris nourries d'une diète de 26% en protéine démontrèrent un plus grand degré d'altérations morphologiques relatives au viellissement que les souris alimentées d'une diète limitée en protéine (4%).
