A total of 78 lactations (25 primiparous and 53 multiparous) in a herd of Murciano–Granadina dairy goats were studied over 3 years. Animals were allocated to two experimental groups: suckling (S) goats were milked once daily until weaning (week 7) and thereafter twice daily; milking (M) goats were milked twice daily from 2 d after parturition. Milk partitioning during milking (machine and machine stripping milk fractions) was recorded every week and the residual milk every 2 weeks. Milk flow rate was studied in 63 lactations on three consecutive days during week 12 or 13. Average machine and machine stripping milk fractions over 210 d were 1·09 and 0·23 1/d for the S and 1·23 and 0·28 1/d for the M group respectively. Machine milk volume and percentage were smaller in the S group during the first 7 weeks of lactation, while the machine stripping fraction was unaffected by group, indicating that this fraction was constant. The average residual milk was 11·1 and 9·2% of total milk in the S and M groups. Goats in their third lactation had the least residual milk (8·9%). Milk flow and total machine milk volume (but not milking time) were affected by parity, second and third lactation goats having higher values. Positive correlations were found between daily milk yield and milk flow characteristics. Residual milk was positively correlated with the machine stripping but not with the machine milk fraction. The results indicated that Murciano–Granadina goats can readily be machine milked, since > 80% of the milk can be obtained without massage or stripping.

Pulsation rates of 40, 60 and 80 cycles/min were combined with pulsator ratios of 50, 60, 70 or 80% in two experiments with different liners. Machine-induced, short-term changes in teat thickness of 14 cows were compared with milk flow rate characteristics and machine strip yields to evaluate the effectiveness of pulsation in relation to liner type. Post-milking teat thickness increased progressively as the b phase of the pulsation cycle was lengthened, and as the d phase was shortened, at all pulsator rates and with either liner. Teat thickness values increased significantly (P < 0·05) when the d phase was <15%. For ratios of 50 and 60%, teat thickness decreased progressively as pulsation rate was increased. At the higher ratios, thickness values were lowest at 60 cycles/min. Pulsation settings that tended to increase teat thickness values also increased both peak milk flow rates and machine strip yields. The influence of liner type on teat thickness changes appeared to be at least as important as the influence of pulsator ratios and greater than the effect of pulsation rate. If so, then international standards for acceptable pulsation characteristics cannot be defined solely in terms of pulsator settings. A limit of ±5% for machine-induced changes in thickness of the teat apex would be an additional guideline for effectiveness of pulsation in relation to both liner type and vacuum level. This could provide a basis for a dynamic test applied to milking cows under field conditions.

Experiments were designed to determine the cisternal milk during machine milking in unfamiliar surroundings, which has previously been shown to inhibit milking-related oxytocin (OT) release and milk ejection. The first experiment was performed with 22 cows in early, mid and late lactation. After cisternal milk was removed, 10 i.u. OT were twice injected intravenously to remove the remaining milk. Total milk yield, cisternal milk yield, cisternal milk fraction (17 and 12% in early and late lactation) and the milk yield obtained in response to the first OT injection significantly decreased from early to late lactation. Cisternal milk yield was similar in front and rear udder halves. However, owing to higher total yield, the cisternal milk fraction was lower in rear than in front halves. Cisternal milk yield and cisternal milk fraction were smaller in primiparous than in older cows. In a further experiment milk distribution in the bovine udder was evaluated after preceding teat stimulation and milk ejection. Teats of 12 cows were manually stimulated for 1 min at 15, 60 or 120 min before milking. Total milk yield was not significantly different with or without teat stimulation. However, cisternal milk yield and fraction were significantly higher in teat stimulated cows than in unstimulated controls but were similar whether cows were stimulated 15, 60 or 120 min before milking.

Ten pregnant heifers were inoculated in both hind udder quarters with a mixture of Actinomyces pyogenes, Fusobacterium necrophorum and Peptostreptococcus indolicus. Development of the experimental mastitis was monitored by sequential clinical and bacteriological examinations, and by blood acute phase protein profiles. Sequential changes in plasma fibrinogen, serum haptoglobin, acid-soluble glycoproteins and α1-proteinase inhibitor activity were analysed and compared with both the clinical and bacteriological findings and the final outcome of the disease after calving. All ten heifers developed moderate to severe clinical mastitis. Four recovered completely, had a normal lactation after calving and exhibited only transient fever and moderate local signs. In six of the heifers the course of the disease was severe, and the inoculated quarters failed to produce milk after calving. The acute phase response of the four heifers that recovered was also significantly milder than that of the other six heifers. Haptoglobin and acid-soluble glycoproteins were most effective in indicating the severity of the infection and predicting the final outcome of the disease. Fibrinogen was a reliable indicator for detecting the presence of bacterial infection in all heifers. α1-Proteinase inhibitor activity was of low diagnostic value in this study.

For six successive milkings, six dairy cows were relocated immediately before milking to an unfamiliar operating theatre, a procedure previously shown to inhibit oxytocin release and milk ejection. Two control milkings were performed in familiar surroundings. After milk flow had ceased, two i.v. injections of 1 i.u. oxytocin were given to remove the remaining milk. Milk flow was recorded continuously and blood samples were taken every minute during milking and 10 min after milking. During the first milking in unfamiliar surroundings, no oxytocin was released. Thereby, only 13% of the total milk yield, the cisternal milk, was available and the alveolar milk fraction could only be removed after injection of oxytocin. During subsequent relocations oxytocin release steadily increased toward the control level, although the timing of oxytocin release remained delayed as compared with controls. However, the milk fraction available before oxytocin injection increased with increasing number of removals, following an asymptotic approach to control levels. The concentrations of β-endorphin, cortisol (and perhaps also of prolactin) gradually declined with the number of times the animal was moved to unfamiliar surroundings, i.e. hormone concentrations gradually adjusted to control level. During milking, concentrations of prolactin and cortisol increased, while β-endorphin concentrations decreased (except for the first relocation). We conclude that milking-related oxytocin release and therefore milk ejection adapted gradually to repeated relocations to unfamiliar surroundings. This adaptation was inversely related to β-endorphin concentrations, so it is possible that oxytocin release was suppressed by high circulating β-endorphin concentrations.

In two separate experiments, the effects of pulsationless milking and milking vacuum on the rate of keratin removal from the teat canal were determined. Sixteen cows were milked with or without pulsation for either a single milking or for eight milkings. Milking without pulsation removed 10% of keratin present before milking, significantly less than milking with pulsation, which removed 32%. After eight milkings (4 d) without pulsation, up to 20% more keratin had accumulated within the teat canal but the rate of keratin regeneration reduced significantly upon return to pulsation milking. In a second experiment, ten cows were milked at 45 or 55 kPa and with or without pulsation. Only the absence of pulsation significantly reduced keratin loss during milking. Keratin loss during milking appears to be controlled by liner compression rather than by the rate of milk flowing through the teat canal. Pulsationless milking may increase penetrability of the teat canal to bacteria by reducing the natural rate of keratin removal during milking and reducing the rate of keratin regeneration.

Thirty-nine multiparous Holstein cows were used to measure the effect of propylene glycol treatment around parturition on milk yield, reproductive efficiency and some hormone and metabolite concentrations. Cows were assigned randomly to control (n = 19) or propylene glycol treated (n = 20) groups. Propylene glycol (300g) was administered directly mixed with the diet from day 10 prior to the expected calving date until parturition (day 0) and orally after dilution in 1 l water on days 3, 6, 9 and 12. Blood samples were collected on days −20, −5, 0, 3, 10, 25 and 50 while milk samples were taken weekly until 13 weeks post partum. Body condition scores, recorded on days −20, 15 and 50, were not affected by propylene glycol administration. Propylene glycol did not significantly affect milk yield or composition but linear somatic cell score measured from the first 13 weeks post partum was reduced by propylene glycol administration (P < 0·01). Moreover, propylene glycol reduced milk urea (−25 mg/l, P < 0·05), especially during the first 9 weeks post partum. Plasma insulin concentrations were similar in both groups during the experiment while insulin-like growth factor I (P < 0·05) and insulin-like growth factor-binding protein 3 (P < 0·001) levels were higher on days 10, 25 and 50 post partum in the propylene glycol group. Propylene glycol administration decreased plasma non-esterified fatty acid concentrations (P < 0·05 to P < 0·01) but increased total cholesterol levels (P < 0·01) after parturition while 3-hydroxy-butyrate levels were unaffected by the treatment. Changes in the hormone and metabolic concentrations after propylene glycol administration in the last few days of gestation and the first week of lactation seem to indicate that energy balance in the treated group was probably more positive than in the control group. There was also evidence that propylene glycol administration prevented fatty liver syndrome and hastened the resumption of oestrous cycles (P < 0·001).

The aim of this study was to describe the results of teat thickness measurement applied routinely in three commercial dairy herds and to evaluate the influence of machine-induced teat thickness changes on intramammary infection and the frequency of new infection. A total of 1018 fore milk samples and the same number of teat apex measurements have been evaluated. Overall, relative teat thickness changes were normally distributed (mean −0·16%, SD 10·15%), while a specific pattern could be observed within herds. Increases in teat thickness of > 5% were significantly associated with infection and new infection (odds ratio > 1), but the association was not significant when teat thickness decreased by more than 5%. When results were classified according to aetiology, analysis showed that coagulase-negative staphylococcal infections were significantly associated with both increases and decreases in teat thickness numerically greater than 5%.

Teat orifice hyperkeratosis, a commonly observed condition in dairy cows, has been considered a consequence of machine milking and the degree of hyperkeratosis may be increased by a poor milking system. A fully illustrated technique is described which uses a scoring system from 0 for a perfect orifice to 5 for an orifice significantly enlarged with extensively protruding fronds of teat duct keratin. A range of scores found in 25 commercial dairy herds is presented. The scores have been averaged for each cow and the markedly skewed distribution corrected by a square root transformation. This scoring procedure allows comparative measures of hyperkeratosis within and between herds. An 8-fold difference (0·17–1·31) in herd average score was found. Within all herds the score increased with lactational age of the animals and peaked, for any lactation, some 3–4 months post partum, declining as the animals dried off. There was no significant relationship between mean somatic cell count and degree of hyperkeratosis at the herd level. This implies that such chronic pathological changes are unlikely to be related to the level of intramammary infection. It appeared that some hyperkeratosis is an obvious and probably natural response to milking and occurs in a significant proportion of animals in all herds although often only to a slight degree. Much more hyperkeratosis may be a measure of the performance and management of the herd. The genetic influence is unknown. Higher yielding cows will score higher as they milk for longer, but generally high scores may reflect consistent and possibly considerable overmilking. Hyperkeratosis may be an indicator of the quality of management and show the level of attention being paid to the welfare of the herd.

The effects of manual teat stimulation compared with stimulation by the liner of a conventional milking machine and the influence of milk ejection on the course of milk flow during the entire milking were investigated. Twelve cows were milked with or without prestimulation on three quarters and milk flow was recorded while intramammary pressure (IMP) was recorded in the left front quarter and blood samples for the determination of oxytocin (OT) were taken at 1 min intervals. OT concentrations increased very similarly in response to manual teat stimulation or liner stimulation and remained elevated during the entire milking procedure. Milk flow curves were generally bimodal without stimulation and not bimodal during milking after prestimulation. Milk yield was not significantly lower in milkings without than with stimulation, whereas machine-on time was prolonged and peak milk flow rate was reduced during milking without stimulation. Time to reach milk flow plateau, time to reach peak flow rate and IMP profiles were similar in both treatments if time was calculated from the start of stimulation. IMP increased similarly in response to manual or liner teat stimulation. In milkings with and without prestimulation IMP did not reach its maximum until the start of milking. In bimodal milk flow curves the second increase of milk flow occurred concomitantly with the start of IMP increase; ejection IMP generally occurred concomitantly with the milk flow plateau. In some cases during milking without prestimulation only small and transient OT release was observed. In response, IMP did not increase to its maximum and increased further in response to a stripping-related OT release. During the course of milking IMP decreased slightly because of the emptying of the three milked quarters. After the end of milking, and with OT concentrations decreasing to the premilking values, IMP again decreased slightly to another plateau, probably owing to myoepithelial relaxation. Thus the course of milk ejection, i.e. the IMP profile, influenced milk removal markedly during early milking.

The transport of lysine by perfused lactating rat mammary tissue has been examined using a rapid, paired-tracer dilution technique. This experimental approach allowed the characteristics of lysine transport across the blood-facing aspect of the mammary epithelium to be studied. The clearance of lysine from the perfusate was influenced by the extracellular lysine concentration in a fashion consistent with the presence of carrier-mediated transport. Replacing extracellular Na+ with N-methyl-D-glucamine had no significant effect on lysine transport. Lysine uptake was inhibited by extracellular leucine and glutamine but not by α-(methylamino)isobutyric acid. Leucine interacted with lysine transport under Na+-free conditions. It appears that the system for cationic acid transport which is situated in the blood-facing aspect of the lactating rat mammary epithelium may also accept neutral amino acids as substrates.

Post-milking measurements of teat skin chapping score, teat thickness and colonization by Staphylococcus aureus were determined for 11·5 d. Three teats on each of twelve Holstein cows, free from Staph, aureus intramammary infections, were immersed in 1 M-NaOH solution to induce teat chapping; the fourth teat served as a control. To achieve different degrees of chapping, one teat per cow received one immersion after each milking for three consecutive milking periods, a second teat received two immersions, and the third teat received one immersion which coincided with the last immersion of the other two teats. All teats were challenged twice with a skim milk broth culture of Staph, aureus (5 × 106 cfu) after the first and second milking following NaOH treatment. Measures were initiated with the milking following the last Staph, aureus challenges. Tissue thickness of the lateral side of the teat (barrel) and colonization by Staph, aureus declined with time. Thickness of the teat end varied more erratically. Teat skin chapping score was positively correlated (P < 0·001) with tissue thickness of the barrel and Staph, aureus colonization. Thickness of the barrel was not significantly correlated with Staph, aureus colonization. Barrel thickness as a covariate had a significant effect (P < 0·05) on teat skin colonization of Staph, aureus, whereas teat skin score and teat end thickness had no significant effect. Thus, thickness of the lateral side of the teat explained the greatest variation in Staph, aureus teat skin colonization in the model tested.

Mammary gland protein metabolism, determined by an arteriovenous difference technique, was monitored in four Holstein-Friesian dairy cows in response to supplemental dietary protein (provided as rumen-protected soyabean meal) during late lactation (weeks 24–30). Each cow was offered two isoenergetic diets composed of grass silage (170 g crude protein/kg dry matter) plus either a low (108 g/kg) or medium (151 g/kg) crude protein concentrate in a single crossover design involving two 21 d periods. On day 21, arteriovenous measurements across the mammary gland were made during a 13 h continuous i.v. infusion of [1-13C]leucine and with frequent (2 hourly) milk sampling during the final 6 h. Although total milk yield was slightly increased (+1 kg/d) by protein supplementation, milk protein yield was not significantly affected. Whole body protein flux (protein synthesis plus oxidation) was not significantly affected by supplementation. Total mammary gland protein synthesis (milk plus non-milk protein) was also not affected by supplementation but on both diets gland synthesis was always greater (by 20–59%) than milk protein output. The fractional oxidation rate of leucine by the mammary gland was significantly increased by protein supplementation (0·047 v. 0·136). Although the enrichment of leucine in secreted milk protein continued to increase, the final value (at 13 h) was 0·94 of the arterial plasma free leucine plateau value (not significantly different), suggesting almost exclusive use of plasma free leucine for milk protein synthesis. Based on current feeding schemes for dairy cattle, a fixed proportion (0·65–0·75) of the additional protein intake (+490 g/d) should have been partitioned into milk protein. Instead, leucine oxidation by the mammary gland was increased. Whether oxidation of other amino acids was also enhanced is unknown but if amino acid oxidation and the ‘additional’ non-milk protein synthesis occurring in the gland are not crucial to milk synthesis, then by reducing such activities improvements in the efficiency of converting absorbed amino acid into milk protein can be achieved.

The effects of bulk milk cell count (BMCC) and stage of lactation on the quality and storage characteristics of UHT milk were investigated. The UHT milk was manufactured in a pilot plant using milk of low BMCC from early and late lactation, and milk of high BMCC from early and late lactation. Upon storage at 20°C, early lactation UHT milk gelled far ahead of late lactation milk. Within each stage of lactation, high BMCC milk tended to gel first. Few differences in the organoleptic properties of the UHT milks were observed. It was apparent that the onset of age gelation may not always be related directly to the level of proteolysis, and that other factors influencing milk composition and the reactions between milk components may play more important roles. At a particular stage of lactation, proteolysis induced by mastitis may hasten the onset of gelation.

The distribution of albendazole (ABZ) and its main metabolites albendazole sulphoxide (ABZSO), albendazole sulphone (ABZS02) and albendazole 2-aminosulphone (NH2ABZSO2) were investigated in bulk milk and milk products after administration of a single oral dose of the drug (12·5 mg/kg) to 80 Laticauda sheep. An analytical method was developed for this investigation from an existing procedure used for the determination of these compounds in plasma and digesta samples. No traces of the parent compound or NH2ABZSO2 were found in milk or milk products, with the exception of the milk collected 36 h after treatment in which 89 μg NH2ABZSO2/kg was detected. Results indicated that ABZ was rapidly oxidized to ABZSO and then to ABZSO2. These metabolites were found at high levels (1–4 mg/kg) in milk collected within 24 h after treatment. Products derived from such milk also contained high concentrations of the two oxidized metabolites, including up to 5 mg ABZSO/kg in Pecorino cheese. Only small quantities of these two metabolites were found in milk collected during the second day after treatment (range 50–500 μg/kg). They were no longer detectable in milk, collected during the third day after dosing, nor were they found in products made from such milk. These findings confirm that the two polar metabolites ABZSO and ABZSO2 were efficiently excreted from the body. Considering that the established maximum residue limit for ovine milk is 100 μg/kg for ABZ plus its metabolites, our results confirmed the appropriateness of the currently prescribed withdrawal time (3 d) after the use of ABZ in lactating sheep. However, considerable levels of ABZSO were detected in milk collected within 24 h after treatment as well as in products and by-products derived from such milk. Owing to the known toxicity of the ABZSO, we stress the need for careful control to ensure adherence to the prescribed withdrawal time.

The effects of heat treatment and subsequent acidification of milk on the distribution of proteins, Ca and Pi, between the serum and micellar phases were examined using ultracentrifugation. After heating milk at 85 °C for 10 min, and storing for 22 h at 4, 20 or 30 °C, there was a marked increase in the concentration of κ-casein in the serum. At 4 and 20 °C there was also slightly more β-casein in the serum from heat-treated milk than in that from the corresponding raw milk. The whey proteins were extensively denatured, and were almost equally distributed between the supernatants and micellar pellets. After storage for 22 h the distribution of Ca and Pi between soluble and colloidal phases in heat-treated milk was similar to that in raw milk. After acidifying heat-treated milk by the addition of glucono-δ-lactone and storing for 22 h at 4, 20 or 30 °C there was progressive solubilization of colloidal calcium phosphate with decreasing pH, and at pH 5·0 almost all of the Ca and Pi was present in the serum. At 20 °C, and even more so at 4 °C, serum concentrations of the individual caseins increased considerably with decreasing pH, reaching maximum levels of about 25 and 40% of the total casein at pH 5·7 and 5·5 respectively, and then decreasing rapidly at lower pH. Compared with raw milk, maximum dissociation in heat-treated milks stored at 4 and 20 °C occurred at higher pH, and the overall levels of dissociation of individual caseins from the micelles were lower. At 30 °C, the concentrations of individual caseins in the serum of heat-treated milk decreased steadily as the pH was reduced, and did not show the slight increase found previously for raw milk. The role of the denatured whey proteins in interacting with κ-casein and in promoting aggregation of the micelles on acidification is discussed.

Four models describing renneting kinetics are evaluated for their ability to describe well documented attributes of the coagulation of casein micelles. The first model is based on a constant flocculation rate parameter. In the second the flocculation rate constant is proportional to the product of the sizes of the aggregating particles. Both models fail to predict proper dependence of rennet coagulation time on enzyme concentration. The third model is based on an energy barrier being reduced in linear proportion to the degree of proteolysis. The enzyme dependency of this model only works when the initial energy barrier is larger than ∼ 50 kBT (where kB is Boltzmann's constant and T the absolute temperature), which does not seem feasible. The fourth model, based on functionality theory, is able to predict proper dependence of rennet coagulation time on enzyme concentration when functionality is ∼ 2.

The effect of Staphylococcus aureus on detachment of bovine mammary epithelial cells in culture was examined. Mammary epithelial cells became detached from fresh monolayers following a 3 h incubation in the presence of Staph. aureus M60. Two different procedures indicated that cell detachment coincided with the S-phase of the cell cycle. The roles of proteinases, toxins and Ca availability in inducing cell detachment were examined. Addition of the proteinase inhibitor phenyl-methylsulphonyl fluoride (1 mM) to the culture medium prevented cell detachment. Addition of a combination of purified staphylococcal proteinases XVI and XVII-B to the culture medium of mammary epithelial cells induced cell detachment in the absence of Staph. aureus. Cell detachment may be caused by a staphylococcal proteinase. However, addition of Ca (10 mM) to the culture medium abolished Staph. aureus-induced cell detachment, despite the fact that proteinase activity was still apparently present. Isogenic mutants of Staph. aureus M60, expressing either ± or β toxins but not both, induced cell detachment, but to a lesser extent than the wild type. Thus, Ca and toxins play some role during cell detachment. Clones established from detached cells that were washed and replated showed the same susceptibility to Staph. aureus-induced cell detachment as the parental cells. This indicated that there is no subclone of mammary epithelial cells more sensitive to this effect.

Measurements of the zeta potential and particle size of casein micelles in skim milk suspensions at natural and lower pH have been made using the technique of electroacoustics. This technique requires no dilution or change of environment of the casein micelles. The zeta potential obtained at natural pH for a commercial skim milk suspension was −18 mV; it became less negative with decreasing pH. The median particle size observed at natural pH for a commercial skim milk suspension was 0·2 εm, in good agreement with previously reported values. The particle size increased as the pH was decreased.

Five ovine αsl-casein variants (A–E) were identified in an Italian population sample using gel electrophoresis at alkaline pH, gel isoelectric focusing, two dimensional gel electrophoresis, and immunoblotting with polyclonal antibodies against αs1-casein. Each casein sample produced two peaks by fast reversed-phase HPLC. Gel isoelectric focusing and electrospray mass spectrometry were used to demonstrate that the first HPLC peak contained the 191 residue αsl-casein molecular species and the second the 199 residue species, in proportions of ∼20:80. Only in the case of the sample containing αsl-casein CE was the method for the separation of the single short and long forms of each variant unsuccessful. Both two dimensional electrophoresis followed by staining with polyclonal antibodies against αsl-casein and electrospray mass spectrometry showed a heterogeneity consistent with that expected from a protein chain with three levels of phosphorylation and two different lengths. However, especially for αsl-caseins D and E, a further uncharacterized heterogeneity was detected.
